GRAIN ROT: How to Burn the Second Floor
Unlocking the Second Floor in GRAIN ROT does not immediately give you access to everything upstairs, because Grain Rot is still blocking part of the area once you finally get through the door. If you have been wandering around looking for another key, switch, or upgrade to remove it, the solution is much simpler: you need to burn the contamination with fire.
A Lantern can give you the fire you need to clear the Grain Rot, although you should move away once the area starts burning because the same flames that remove the contamination can also damage your Vessel. After the Rot is gone, you can continue following the Second Floor objectives and eventually reach the new progression systems upstairs.

To burn the Second Floor in GRAIN ROT, you need to create fire around the Grain Rot contamination blocking your progression upstairs. Once the Second Floor has been unlocked, head into the newly opened area and look for the Rot preventing you from continuing, then use a source of fire to burn it away.
This is one of those progression steps that can seem more complicated than it actually is because most of the major obstacles leading up to this point involve finding an item and returning it somewhere. The Grain Rot is different. You do not need another key or some specific permanent upgrade to get through it; if the contamination is blocking where you need to go, burn it.
The Second Floor only becomes properly useful once you start clearing these blockers, so there is not much reason to unlock the area and immediately head back into another normal Deep Layers run. Deal with the Grain Rot first, then continue exploring upstairs while the new objectives are available.
How to Use a Lantern for the Grain Rot
A Lantern is useful here because it gives you a source of fire when the Grain Rot is stopping your progress. If you reach the Second Floor without another way to deal with the contamination, bring a Lantern upstairs and use its fire against the Rot rather than continuing to search for an interaction prompt that is not going to solve the problem.
I would not start stockpiling Lanterns just because they become useful for this objective, though. Their value here comes from solving a specific blocker, so if fire is the only thing currently preventing you from continuing upstairs, get what you need for that problem and save the rest of your resources for the upgrades that become available afterward.
The important part is the fire itself. Once the Grain Rot has started burning, give it enough room to clear instead of staying directly on top of the flames.
Avoid Taking Damage From the Fire
Fire removes the Grain Rot, but it can also hurt your Vessel, which means clearing the Second Floor can become unnecessarily expensive if you stand inside the burning area while waiting for the contamination to disappear. Start the fire where the Rot is blocking you, then move back rather than treating the flames like a safe environmental effect.
If you do take meaningful damage while clearing it, recover before immediately forcing the next part of the upstairs progression. There is no benefit to getting through the Rot only to enter the next objective with a damaged Vessel, especially when the Second Floor starts introducing systems meant to prepare you for harder runs rather than simply sending you straight back downstairs.
You also do not need to turn the entire process into one prolonged burn. The goal is simply to remove the contamination that is actually blocking progression, so create the fire where you need it, stay out of the dangerous area yourself, and continue once the path is usable.
What to Do After Burning the Second Floor
Once the Grain Rot has been cleared, keep following the objectives on the Second Floor because burning the contamination is only the first part of reclaiming the upstairs area. One of the major progression points that follows is finding Tink, an engineer and mechanic who introduces the Upgrader and gives you another way to improve weapons you already want to keep using.
The Second Floor also introduces new permanent upgrade choices, including Critical Hit Chance, Stamina Recovery, and Sprint Speed, so this is where your resources start having more competing uses. Rather than dumping everything into the first new option you see, it makes more sense to get through the Rot first and see what problem your next run actually needs you to solve.
If you are only stuck on the initial Second Floor blocker, though, you do not need to worry about any of that yet. Bring a source of fire upstairs, burn away the Grain Rot blocking your route, stay clear of the flames while they do the work, and then continue following the Second Floor objectives once the contamination is gone.
