To get Curse in Vampire Crawlers, beat Weeny Bridge, then buy 1 rank of the Curse Power Up from the village shop once it unlocks.
That 1 rank is enough for the Lama Ladonna requirement, since she needs a completed dungeon with 10% or more Curse. After buying the Power Up, start a dungeon, clear it with Curse active, and the requirement should be covered.

This is mostly a patience check. Clear the right stage, buy the rank, finish a dungeon.
Can Curse Come From A Card Too
Curse can also show up later through a yellow card that destroys itself after being used. That can help if it appears, but it is not the clean route for Lama Ladonna.
The shop Power Up is better because it gives a straightforward way to hit the Curse requirement before starting the dungeon. Waiting for the right card during a run is less reliable and makes the unlock feel more annoying than it needs to be.
What Curse Does
Curse makes enemies stronger and increases XP gain. It is a tradeoff stat, not a free upgrade.
That means Curse can be useful once a build is strong enough to handle tougher enemies, but it can also make weak runs worse fast. For Lama Ladonna, the goal is not to make the run spicy for no reason. The goal is just to get 10% Curse active and finish the dungeon.
